The efficiency of enzymatic conversion of glucose into glucose-6-phosphate in fish fed carbohydrates is still a matter of debate. This study reports the first ever identification of hexokinase-family members at a molecular level in fish. The deduced amino acid sequences of six cDNA clones allowed their assignement either to hexokinase IV (glucokinase) for rainbow trout (Oncorhynchus mykiss), common carp (Cyprinus carpio) and gilthead seabream (Sparus aurata) or to hexokinase I/II for the common carp (two clones) and the gilthead seabream, based on their similarities to known members of the HK family. The evolutionary relationships among these sequences were investigated by phylogenetic analysis. These hexokinase sequences constitute potential molecular tools for analysis of interspecific differences in dietary carbohydrate utilization in fish.  相似文献

In this study, the sorption characteristics of U(VI) onto eucalyptus biochar as a function of various operating parameters such as solution pH, initial metal ion concentration, contact time and ionic strength of the medium are reported. Biochar was characterised using various techniques such as CHNS element analysis, Fourier transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA), scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDS) and X-ray photoelectron spectroscopy (XPS). SEM analysis showed the presence of micro- and macropores in the sorbent, and FTIR spectra confirmed the presence of functional groups like carboxylic (?COOH), hydroxyls(?OH), carbonyls(–C=O), etc. Maximum sorption of about 95% is found to occur in the pH range of 5 to 6. U(VI) sorption onto biochar reached equilibrium within 20 min at pH 5.5. The kinetic data were analysed using both pseudo-first-order and pseudo-second-order kinetic models, and the latter is found to be more appropriate to explain the observed kinetics. The equilibrium data were correlated with Langmuir and Freundlich models, and the maximum monolayer adsorption capacity obtained from the Langmuir model was 27.2 mg/g at 293 K. From EDS, FTIR and XPS measurements, it is found that the sorption process involves chemical interaction between the U(VI) and the surface functional groups on the adsorbent. Efficient removal of low level of uranium from ammonium diuranate supernatant demonstrates its utility as sorbent for waste water treatment.  相似文献

In the present study, seven fertilizer treatments [T1, 50% NPK; T2, 100% NPK (Recommended dose of fertilizer, 200–65.4–124.5 kg N-P-K ha?1); T3, 150% NPK; T4, 100% PK; T5, 100% NK; T6, 100% NP and T7, control (zero NPK)] with four replications were assessed in the new alluvial soil zone (Entisols) of West Bengal, India. The objectives of the study were to generate information on potato productivity, profitability, indigenous nutrient supply and net gain/loss of NPK in post-harvest soil. Plants grown under higher NPK supply resulted in higher tuber yield and there were significant (p ≤ 0.05) reductions in total yield with nutrient omissions. Nutrient?limited yields were 19.78, 2.83 and 1.77 t ha?1 for N, P and K, considering total tuber yield (28.24 t ha?1) obtained under 100% NPK as targeted yield. Indigenous nutrient supply of N, P and K were estimated at 24.1, 22.34 and 110.22 kg ha?1, respectively that indicates higher K?supplying capacity of experimental soil as compared to N and P. Net income (US$1349 ha?1 year?1) and B:C ratio (1.91) was highest with 100% NPK, and further addition of NPK (150%) resulted in decrease on net return (US$1193 ha?1 year?1) and B:C ratio (1.73).  相似文献

Two basal diets M0 and V0 were formulated with marine and plant based ingredient composition. Seven experimental diets were prepared from the two basal diets namely M0, M100, V0, V30, V60, V100 and V150 by incorporating different levels of a micromineral premix (Cu, Fe, Mn, Se and Zn). Triplicate groups of rainbow trout (initial weight: 20 g) reared at 17°C were fed one of each diet to apparent visual satiation over 12 weeks. Among the V diet fed fish, growth and feed intake exhibited maximal response at V60 level of premix inclusion; Apparent availability coefficient of Fe, Cu and Zn decreased linearly with increasing level of premix whereas apparent availability coefficient of Mn and Se was unaffected. The available dietary concentration in basal V0 diet was for Fe, 20.6; Cu, 2.8; Mn, 6.5; Zn, 17.3 and Se, 0.195 (in mg/kg DM) and in the M0 diet for Fe, 63.3; Cu, 5.2; Mn, 2.9; Zn, 35.2 and Se, 0.87 (in mg/kg DM). In reference to NRC (Nutrient requirements of fish and shrimp. Washington, DC: National Research Council, The National Academies Press, 2011) recommendations, the V0 basal diet accounted for 34.3%, 92.9%, 53.9%, 115% and 130.2% and the contribution from M0 diet for 105.5%, 173.3%, 24.2%, 234.7% and 580% of the minimal dietary inclusion levels of Fe, Cu, Mn, Zn and Se to rainbow trout, respectively. However, data on whole body mineral contents showed that normal levels were maintained only for Cu and Mn through supply from basal V0 diet. For Zn and Se, available supply even from the highest supplemented diet (V150) was not sufficient to maintain normal body mineral levels of rainbow trout in the present study. On the whole, optimal dietary inclusion levels of microminerals are altered while using fishmeal‐free diets for rainbow trout.  相似文献

ABSTRACT

This case study assessed local food stakeholders' use of Facebook and Twitter to support interaction and build their networks of innovation in Ontario. Data were collected using Netlytic ? an online data mining tool from the social media platforms ? and key informant interviews. Findings revealed that stakeholders could be more innovative in their use of social media, but they would be unlikely to do so, without tapping into beneficial interactions of weak ties, as well as fostering strong ties. They also need to utilize the high brokerage role of key facilitating organizations and develop a social media strategy by integrating both ‘online’ and ‘offline’ interactions.  相似文献

The substance secreted by mussels, also known as nature’s glue, is a type of liquid protein that hardens rapidly into a solid water-resistant adhesive material. While in seawater or saline conditions, mussels can adhere to all types of surfaces, sustaining its bonds via mussel adhesive proteins (MAPs), a group of proteins containing 3,4-dihydroxyphenylalanine (DOPA) and catecholic amino acid. Several aspects of this adhesion process have inspired the development of various types of synthetic materials for biomedical applications. Further, there is an urgent need to utilize biologically inspired strategies to develop new biocompatible materials for medical applications. Consequently, many researchers have recently reported bio-inspired techniques and materials that show results similar to or better than those shown by MAPs for a range of medical applications. However, the susceptibility to oxidation of 3,4-dihydroxyphenylalanine poses major challenges with regard to the practical translation of mussel adhesion. In this review, various strategies are discussed to provide an option for DOPA/metal ion chelation and to compensate for the limitations imposed by facile 3,4-dihydroxyphenylalanine autoxidation. We discuss the anti-proliferative, anti-inflammatory, anti-microbial activity, and adhesive behaviors of mussel bio-products and mussel-inspired materials (MIMs) that make them attractive for synthetic adaptation. The development of biologically inspired adhesive interfaces, bioactive mussel products, MIMs, and arising areas of research leading to biomedical applications are considered in this review.  相似文献

The effect of high pressure (HP) treatment (at 100, 270, and 435 MPa for 5 min at 25°C) on microbiological (total viable count and total psychrotrophic count), physical (color, texture, and drip loss), and microstructural characteristics of black tiger shrimp (Penaeus monodon) during storage at 2°C for 35 days was investigated. Pressure treatment increased drip loss, maintained low microorganisms level, imparted cooked appearance to the muscle, and resulted in improved texture. Results of scanning electron microscopy revealed more compact structure in treated samples, confirming the results of texture profile analysis. Pressure treatment of 435 MPa was most effective in preserving the quality of shrimp.  相似文献

Since germination has been found to increase the nutritive qualities of grains, proso millet, a minor millet, was allowed to germinate for 1–7 days and analysed for protein fractions, free amino acids, lysine, tryptophan, methionine, non-protein nitrogen, starch, and sugars. Overnight soaking and germination up to 7 days significantly increased the free amino acids and total sugars while the content of dry weight and starch decreased. Increases in albumin and globulin and large decreases in prolamin accompanied sprouting. The percent protein in germinated grains was higher than in the initial grain as a result of dry matter loss during germination. Further, there was an increase in lysine, tryptophan and non-protein nitrogen contents during germination. No change was noticed in methionine content.  相似文献

Cell-cell contacts are fundamental to multicellular organisms and are subject to exquisite levels of control. Human RPTPmu is a type IIB receptor protein tyrosine phosphatase that both forms an adhesive contact itself and is involved in regulating adhesion by dephosphorylating components of cadherin-catenin complexes. Here we describe a 3.1 angstrom crystal structure of the RPTPmu ectodomain that forms a homophilic trans (antiparallel) dimer with an extended and rigid architecture, matching the dimensions of adherens junctions. Cell surface expression of deletion constructs induces intercellular spacings that correlate with the ectodomain length. These data suggest that the RPTPmu ectodomain acts as a distance gauge and plays a key regulatory function, locking the phosphatase to its appropriate functional location.  相似文献

Glucose plays a key role as energy source in the majority of mammals, but its importance in fish appears limited. Until now, the physiological basis for such apparent glucose intolerance in fish has not been fully understood. A distinct regulation of hepatic glucose utilization (glycolysis) and production (gluconeogenesis) may be advanced to explain the relative inability of fish to efficiently utilize dietary glucose. We summarize here information regarding the nutritional regulation of key enzymes involved in glycolysis (hexokinases, 6-phosphofructo-1-kinase and pyruvate kinase) and gluconeogenesis (phosphoenolpyruvate carboxykinase, fructose-1,6-bisphosphatase and glucose-6-phosphatase) pathways as well as that of the bifunctional enzyme 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase. The effect of dietary carbohydrate level and source on the activities and gene expression of the mentioned key enzymes is also discussed. Overall, data strongly suggest that the liver of most fish species is apparently capable of regulating glucose storage. The persistent high level of endogenous glucose production independent of carbohydrate intake level may lead to a putative competition between exogenous (dietary) glucose and endogenous glucose as the source of energy, which may explain the poor dietary carbohydrate utilization in fish.  相似文献
